Agonist activity against AhR

Exploratory Research

Measured value Ratio to control
Animal species Others
Indication of the result The ratio of the strength of AhR activation under test drug treatment to that for the control condition (water or DMSO) is shown. Values larger than 1 indicate the activation of the receptor by test drug.
Target diseases of assay Endcrine & Metabolic systems
Assay level Cell-based
Protocol of assay 1) HepG2, a human hepatoma-derived cell line, was transfected with XRE-luciferase reporter plasmid and beta-galactosidase expression plasmid by calcium phosphate method. 2) Test drug solution was added, and the cells were incubated for 48 h. 3) The cells were lysed, and luciferase and beta-galactosidase activities were measured. 4) The strength of AhR activation was calculated by correcting the value of luciferase activity using the value of beta-galactosidase activity.
