Indication of the result The ratio of the amount of viable cells under test drug treatment to that for the control condition (0.1% DMSO or medium only) is shown. Values less than 100 indicate cytotoxic effect on the HL60 cells.

Protocol of assay 1) HL60 cells were seeded at 10000 cells/well on 96-well plates with 100 μL of a culture medium including test drug solution. 2) The cells were incubated for 72 h at 37 °C with 5 % CO2. 3) 10 μL of the WST-8 reagent (Dojindo, Kumamoto, Japan) was added into each well followed by incubation for 3 h at 37 °C. 4) The amount of viable cells was measured by the absorbance at 450 nm using a SpectraMax PLUS 384 microplate reader (Molecular Devices, Sunnyvale, CA, USA).
