Indication of the result The ratio (%) of the amount of snRNP under test drug treatment to that for the control condition (DMSO or pure water) is shown. Values less than 100 indicate reduction of cellular snRNP level.

Protocol of assay 1) The 293T cells stably expressing the snRNP reporter were seeded in 96-well plates and treated with test drug solutions. The culture volume was 200 μL. 2) The cells were incubated for 4 h at 37 °C. 3) Culture medium was removed, and 150 μL of lysis buffer was added. 4) Cells were lysed by pipetting, and 120 μL of the lysate was mixed with 50 μL of substrate solution Steady-Glo in white 96-well plates. 5) The cellular snRNP level was measured by the luminescence intensity. The values were corrected based on the location of each well in a plate and were averaged over three measurements.
