Protocol of assay 1) BV-2 cells were seeded at a density of 7.5×10^4 cells/well in 96-well plates for Griess assay. Test drug solution was added to the wells, and the cells were incubated for 30 min. 2) LPS (0.1 μg/mL) was added to the medium, and the cells were incubated for 24 h. 3) Cell-free supernatants were collected and mixed with an equal volume of Griess reagent (1 % sulfanilamide, 0.1 % naphthylethylenediamine dihydrochloride, and 2.5 % phosphoric acid). 4) Absorbance was measured at 570 nm with a microplate reader (Bio-Rad, Hercules, CA, USA) and nitrite concentrations were determined with reference to a calibration curve prepared with sodium nitrite standards.
