Protocol of assay 1) 3 mL of the test substance solution (3 mg/mL or 3 mM) and 27 μL of buffer were mixed in a 384-well plate. 2) 10 mL of substrate (BOC-GRR-MCA) and 17 mL of enzyme (NS2B gly NS3) were added. The mixture was left at room temperature for 20 min. 3) The NS2B-NS3 enzyme activity was measured by fluorescence intensity (excitation wavelength 360 nm, fluorescence wavelength 440 nm).
