Indication of the result Inhibition (%) = 100 x [1-(A-B)/(C-B)]. A, B, C mean absorbance at 670 nm of sample with test drug, background, and control (no drug), respectively. Positive values indicate inhibitory effect on hydrogen sulfide-producing enzyme activity.

Protocol of assay 1) Reaction mixture was incubated for 40 min at 25 °C. The composition of 100 μl of reaction mixuture was 30 mM Hepes pH7.5, 1 μM Pyridoxal 5'-phosphate, 1 mM Dithioerythritol, 150 μM L-Cysteine, 0.03 μM Enzyme, 0.01 % (v/v) Triton X-100, 1.0 % DMSO, and 100 μM compound (or 0.1 mg/ml crude drug extract). 2) 20 μl of termination solution was added to the mixtures. The resulting solution was incubated at room temperature for 30 min. The composition of the termination solution was 10 mM N,N-Dimethyl-p-phenylenediamine dihydrochloride, 15 mM FeCl3, and 4.2 M HCl. 3) The hydrogen sulfide-producing enzyme activity was measured by the absorbance at 670 nm.
