Protocol of assay 1) HeLa cells were cultured in a 96-well plate. 2) The medium was replaced with fresh medium, and fluorescent substrates and DMSO solution of test drug were added. Cells were incubated. 3) The plates were washed by PBS three times. The enzyme activity was measured by the fluorescence intensity with a multi-mode microplate reader.
