Indication of the result The rate of decrease (%) in the enzyme activity of prolyl tripeptidyl peptidase under test drug treatment against the control condition without test drug is shown. Positive values indicate inhibition of the enzyme activity.

Protocol of assay 1) The reaction mixture consisted of 10 μl of test drug solution, 10 μl of enzyme solution, 10 μl of a 2 mM solution of substrates (Gly-Ala-Pro-βNA), and 70 μl of 20 mM Tris-HCl buffer (pH 7.0). The mixture was incubated for 10 min at 37 °C. 2) The reaction was stopped by adding 50 μl of Fast Garnet GBC (1 mg/ml) solution containing 10% Triton X-100 in 1 M sodium acetate buffer (pH 4.0). The mixture was incubated for 20 min. 3) The amount of azo-dye generated by the enzyme reaction catalyzed by prolyl tripeptidyl aminopeptidase was measured by the absorbance at 550 nm.
