Indication of the result The ratio (%) of the promoter activity of Piwil1 (Miwi) gene under test drug treatment to that for the control condition with water is shown. Values less than 100 indicate the inhibition of the promoter activity of Piwil1 gene.

Protocol of assay 1) 293LTV cells were transfected with MIWI promoter-Gluc and Cluc-expressing plasmids and cultured for 24 h at 37 °C. 2) The cells were cultured with test drug for 24 h at 37 °C. 3) 5 μl of sample solution from the cells was mixed with 5 μl of assay buffer according to the NEB protocol. 4) Luciferase activity was measured by the fluorescence intensity. 5) The promoter activity of Piwil1 gene was calculated by normalizing the value of Gluc using the value of of Cluc.
