Indication of the result The ratio (%) of hydrolysis activity of hyaluronidase under test drug treatment to that for the control condition without test drug is shown. Values less than 100 indicate inhibition of hydrolysis activity of hyaluronidase.

Protocol of assay 1) 5 μl of test drug, 12.5 μl of buffer, and 27.5 μl hyarulonic acid (HA) solution as a substrate were mixed in a microtube. 2) 10 μl of bovine testicular hyaluronidase (BTH) was added, and the reaction mixture was incubated for 1 h at 37 °C. 50 μl of the reaction mixture contained sodium acetate buffer (final concentration 100 mM, pH 5.0)-150 mM NaCl, HA (Mr=80,000) 10 μg, and hyaluronidase 10 μg. 3) The reaction was stopped by boiling for 5 min. 4) The reaction products were analyzed by HPLC monitored at 215 nm. Hydrolysis activity of hyaluronidase was assessed according to the peak areas of HA tetra- and hexasaccharides (HA4 and HA6, respectively), which were major final products.
