Stimulatory effect on neurite outgrowth

Exploratory Research

Measured value Ratio to control (%)
Animal species Rat
Indication of the result The ratio (%) of the total length of neurites per cell under test drug treatment to that for the control condition with water is shown. Values greater than 100 mean neurite outgrowth.
Target diseases of assay Brain, Nervous system
Assay level Cell-based
Protocol of assay 1) PC12 cells (0.1 × 10^5 cells/ml) were seeded on collagen coated 12-well plates and cultured for 24 h. 2) Cells were treated with test drugs (0.1 mg/ml). 3) After 3 or 6 days, PC12 cells were photographed using an inverted microscope and phase-contrast objectives. Images of two fields per well were taken with 40–50 cells per field. 4) The length of neurites was measured for 20 independent cells per field using Image J 1.44 software.
