Indication of the result The ratio of the level of YAP1 activity under test drug treatment to that for the control condition (DMSO-treated or water-treated) is shown. Values greater than 1 indicate stimulatory effect on the transcriptional regulator activity of YAP1.

Protocol of assay 1) The ARPE-19 YAP1 reporter cells (Maruyama et al. (2018) Mol. Cancer Res. 16 197-211.), in which the activity of YAP1-TEAD transcription factor complex is measurable via H2B-mCherry fluorescence intensity, were seeded into 96-well plates. 2) Each natural product-derived compound was added at the final concentration of 10 µM. Also, each crude drug or Kampo formula extract was added at the final concentration of 100 µg/ml. 3) After 3 d, The YAP1 activity was measured by the H2B-mCherry fluorescence intensity in each well by using Thermo Fisher Arrayscan VTI.
